The complex hexaploid wheat genome offers many challenges for genomics research. Expressed sequence tags facilitate the analysis of gene-coding regions and provide a rich source of molecular markers for mapping and comparison with model organisms. The objectives of this study were to construct a high-density EST chromosome bin map of wheat homoeologous group 2 chromosomes to determine the distribution of ESTs, construct a consensus map of group 2 ESTs, investigate synteny, examine patterns of duplication, and assess the colinearity with rice (...) of ESTs assigned to the group 2 consensus bin map. A total of 2600 loci generated from 1110 ESTs were mapped to group 2 chromosomes by Southern hybridization onto wheat aneuploid chromosome and deletion stocks. A consensus map was constructed of 552 ESTs mapping to more than one group 2 chromosome. Regions of high gene density in distal bins and low gene density in proximal bins were found. Two interstitial gene-rich islands flanked by relatively gene-poor regions on both the short and long arms and having good synteny with rice were discovered. The map locations of two ESTs indicated the possible presence of a small pericentric inversion on chromosome 2B. Wheat chromosome group 2 was shown to share syntenous blocks with rice chromosomes 4 and 7. (shrink)

We describe a set of two computer‐implemented models that solve physics problems in ways characteristic of more and less competent human solvers. The main features accounting for different competences are differences in strategy for selecting physics principles, and differences in the degree of automation in the process of applying a single principle. The models provide a good account of the order in which principles are applied by human solvers working problems in kinematics and dynamics. A total of 1918 loci, detected by the hybridization of 938 expressed sequence tag unigenes from 26 Triticeae cDNA libraries, were mapped to wheat homoeologous group 4 chromosomes using a set of deletion, ditelosomic, and nulli-tetrasomic lines. The 1918 EST loci were not distributed uniformly among the three group 4 chromosomes; 41, 28, and 31% mapped to chromosomes 4A, 4B, and 4D, respectively. This pattern is in contrast to the cumulative results of EST mapping in all homoeologous groups, as reported (...) elsewhere, that found the highest proportion of loci mapped to the B genome. Sixty-five percent of these 1918 loci mapped to the long arms of homoeologous group 4 chromosomes, while 35% mapped to the short arms. The distal regions of chromosome arms showed higher numbers of loci than the proximal regions, with the exception of 4DL. This study confirmed the complex structure of chromosome 4A that contains two reciprocal translocations and two inversions, previously identified. An additional inversion in the centromeric region of 4A was revealed. A consensus map for homoeologous group 4 was developed from 119 ESTs unique to group 4. Forty-nine percent of these ESTs were found to be homoologous to sequences on rice chromosome 3, 12% had matches with sequences on other rice chromosomes, and 39% had no matches with rice sequences at all. Limited homology was found between wheat ESTs on homoeologous group 4 and the Arabidopsis genome. Forty-two percent of the homoeologous group 4 ESTs could be classified into functional categories on the basis of blastX searches against all protein databases. (shrink)

Cyclin-dependent kinase 5 has been implicated in Alzheimer's disease pathogenesis. Here, we demonstrate that overexpression of p25, an activator of cdk5, led to increased levels of BACE1 mRNA and protein in vitro and in vivo. A p25/cdk5 responsive region containing multiple sites for signal transducer and activator of transcription was identified in the BACE1 promoter. STAT3 interacts with the BACE1 promoter, and p25-overexpressing mice had elevated levels of pSTAT3 and BACE1, whereas cdk5-deficient mice had reduced levels. Furthermore, mice with a (...) targeted mutation in the STAT3 cdk5 responsive site had lower levels of BACE1. Increased BACE levels in p25 overexpressing mice correlated with enhanced amyloidogenic processing that could be reversed by a cdk5 inhibitor. These data demonstrate a pathway by which p25/cdk5 increases the amyloidogenic processing of APP through STAT3-mediated transcriptional control of BACE1 that could have implications for AD pathogenesis. The DAMA project is an observatory for rare processes and it is operative deep underground at the Gran Sasso National Laboratory of the I.N.F.N. In particular, the DAMA/LIBRA (Large sodium Iodide Bulk for RAre processes) set-up consists of highly radiopure NaI(Tl) detectors for a total sensitive exposed mass of ≃250 kg. Recent results, obtained by this set-up by exploiting the model independent annual modulation signature of Dark Matter (DM) particles, have confirmed and improved those obtained by the former DAMA/NaI experiment. (...) A model independent evidence for the presence of Dark Matter particles in the galactic halo is cumulatively obtained at 8.2 σ C.L. No systematics or side reactions able to account for the measured modulation amplitude and to contemporaneously satisfy all the many specific requirements of the signature have been found or suggested by anyone over more than a decade. An example of one of the many possible model dependent corollary quests for the candidate particles and for the related astrophysical, nuclear and particle physics scenarios is presented considering the whole cumulative exposure. The participation of vulnerable patients in clinical research poses apparent ethical dilemmas. Depending on the nature of the vulnerability, their participation may challenge the ethical principles of autonomy, non-maleficence, or justice. On the other hand, non-participation may preclude the building of a knowledge base that is a prerequisite for defining the optimal clinical management of vulnerable patients. Such clinical uncertainty may also incur substantial economic costs. We present the participation of pre-menopausal women with atrial fibrillation in trials of novel oral (...) anticoagulant drugs as a case study. Due to their non-participation in pivotal trials, it is uncertain whether for them, the risks that are associated with these drugs are outweighed by the advantages compared with conventional treatment. We addressed the question whether research of this new class of drugs in this subgroup would be appropriate from both, an ethical as well an economic perspective. We used the method of specifying norms as a wider framework to resolve the apparent ethical dilemma, while incorporating the question whether research of oral anticoagulants in premenopausal women with atrial fibrillation can be justified on economic grounds. For the latter, the results of a value-of-information analysis were used. Further clinical research on NOACs in premenopausal women with atrial fibrillation can be justified on both, ethical and economic grounds. Addressing apparent ethical dilemmas by invoking a method such as specifying norms can improve the quality of public practical reasoning. As such, the method should also prove valuable to committees that have formally been granted the authority to review trial protocols and proposals for scientific research. (shrink)

Searches for non-paulian nuclear processes, i.e. processes normally forbidden by the Pauli–Exclusion–Principle (PEP) with highly radiopure NaI(Tl) scintillators allow the test of this fundamental principle with high sensitivity. Status and perspectives are addressed.

This report describes the rationale, approaches, organization, and resource development leading to a large-scale deletion bin map of the hexaploid wheat genome. Accompanying reports in this issue detail results from chromosome bin-mapping of expressed sequence tags representing genes onto the seven homoeologous chromosome groups and a global analysis of the entire mapped wheat EST data set. Among the resources developed were the first extensive public wheat EST collection. Described are protocols for sequencing, sequence processing, EST nomenclature, and the assembly of (...) ESTs into contigs. These contigs plus singletons were used for selection of distinct sequence motif unigenes. Selected ESTs were rearrayed, validated by 5′ and 3′ sequencing, and amplified for probing a series of wheat aneuploid and deletion stocks. Images and data for all Southern hybridizations were deposited in databases and were used by the coordinators for each of the seven homoeologous chromosome groups to validate the mapping results. Results from this project have established the foundation for future developments in wheat genomics. (shrink)

Guilders 9.90.) Contemporary students of medieval philosophy, especially those influenced by the writings of Gilson, usually view Augustine as primarily an essentialist in metaphysics, while Aquinas is viewed as some sort of existentialist. This is taken to mean that, whereas Augustine seems to identify being with essence (...) or wha~ a thing is, Aquinas distinguishes between what a thing is and its act of existence. The major contribution of Aquinas to later philosophy is often depicted as this distinction between essence and existence. Perhaps the major interest and value of Professor Anderson's essay is that it contests this currently predominant opinion of the essentialist character of Augustinian metaphysics and calls for a re-evaluation of the supposedly revohi~ionary character of the Thomistic concentration upon the act of existence. It is quite evident that Augustine broke with Plotinus when he asserted that the first principle of all things was God, being itself (ipsum esse), rather than the One, which transcends being. The controversy arises over Augustine's view of the nature of being (esse). For Gilson the notion of being remained for Augustine Neoplatonic, while for Anderson Augustine introduced an existential aspect to the notion of being lacking in Neoplatonic thought. Citing the same texts for support Gilson maintains that Augustine interprets the name of God given in Exodus as "I am he who never changes," while Anderson maintains that Augustine interprets Exodus as asserting that God is being, in the sense of existence, itself. The major point of disagreement between Gilson and Anderson is not whether Augustine regarded being as pre~ dominant, but how Augustine interpreted the notion of being. In some texts Augustine maintsSns that being and immutability mean the same thing, in others he argues that a thing truly is 'because it is immutable, and in yet others he argues that a thing is immutable because it truly is. The first opinion seems to be the one frequently favored as an interpretation of Augustine by Gilson. The second is usually regarded as Neoplatonic. The third is the opinion which Professor Anderson argues was most accurately Augustine's. According to Professor Anderson the first two opinions reflect for Augustine a merely factual connection between being and immutability, whereas the third reflects a metaphysical entailment. I should like to mention three points in Professor Anderson's discussion which seem to me weak. Firstly, as Augustine remarks that being is the name for unchangeableness we cannot take this as asserting a merely factual relationship. Secondly, to say that 'being' and 'immutability ~mean the same thing to Augustine or that being and immutability are identical to him does not necessitate, as Professor Andersen claims, denying the primacy of being. If 'being' and 'immutability' mean the same thing, then it is very peculiar to ask which is primary. If it is remarked that 'triangleI means the same thing as 'three sided figure', what sense does it make to ask which notion is primary? I suspect that Professor Anderson's contention makes sense only if one believes that there is a difference in the meanings of 'being' and 'immutability ', but whether or not there was such a difference for Augustine is precisely the point at issue. Professor Anderson asserts that it would be a "metaphysical inconsistency" to claim that God is both being and immutability, but it is not clear why such a claim would be inconsistent if the two terms in fact mean the same thing (p. 30). Thirdly, if, as is in fact the case, Augustine's remarks on the relationship between immutability and being are inconsistent perhaps we should accept the possibility that Augustine himself never came to a clear view on this matter rather than attempt to surmise what Augustine "must have meant." Of course, such inconsistency in Augustine's writings would tend to support the thesis of Gilson that Augustine has difficulties reconciling his theology and metaphysics. For the (...) problems constructed and dealt with in this way emerge from some earlier philosophical clime: from the worlds of Meinong's Investigations Into the Theory of Objects and Russell's Lectures on Logical Atomism, where the talk is of "elementary propositions," "The Ideal Language," "bare particulars," "denoting," what "subsists" as distinct from what "exists," and so on. If any of the writers are aware of the contributions made to philosophy by the later Wittgenstein (mentioned briefly only once in the text, p. 89), Wisdom (not referred to at all), Austin (who appears in a footnote as a translator of Frege), Ryle (mentioned once), and Strawson, this awareness is suppressed with a ruthlessness that borders on perversity. Viewing experience from this wide perspective, and including in it what the Germans have called Erlebnis in order to do justice to the empirical aspects of existentialism as well as of phenomenology, these scholars have performed a basic service for the sciences and for philosophy. In addition to the more traditional distinctions between cognitive, moral, aesthetic, and religious experience, there are illuminating accounts of geological and biological empiricism (relating the terrestrial sciences to nuclear and cosmic sciences), and of experience englobante as distinguished from experience objective. Though each essay is illuminating, those on Existential Philosophy: its historical meaning (1944) and The Theological Significance o/Existentialism and Psychoanalysis (1955) may be called brilliant. Part III contains comparisons of theological thought in Germany and America (in which the author gives some personal reminiscences), and of religion in America and Russia, together with An Evaluation o/Martin Buber (1948). The book concludes with Part IV: Communicating the Christian Message: a Question to Christian Ministers and Teachers (1952), which appears to be relevant to Christians in general. It may be said that it has been a privilege for America to have received among the intellectual refugees of the "catastrophe" of 1933 this powerful German thinker, just as it must have bcen a privilege for Dr. Tillich to be able to elaborate his thought in a country where it received such great response in thoughtful circles. One recognizes behind this thinking the essential continental problems of the relation between philosophy of religion and theology, between religion and culture, between personal existence and industrial society: questions becoming American problems as well. In all his work, the role of religion is positively appreciated, and an appeal is made to a religious "courage to be," reminding man in his estranged life situation of his essential nature and his final destiny. It is not with the deus est esse, neither the certitudo ex se ipsis, nor the ultimate concern, nor the unconditional, that we should like to voice here some questions with regard to the author's views. Rather it is with two seemingly minor questions. Accepting his circumscription of religion as "the dimension of depth," we would be interested to see if within or beside a Theology o] culture, a Theology o] religion would also be possible, having as its subject religious expressions under and outside the authority of the Gospel. A second question arises from the author's statement: "Self is good, self-affirmation is good, self-acceptance is good, but selfishness is bad because it prevents both self-affirmation and self-acceptance" (p. 145). Although this phrase may have its particular context in Moralisms and Morality (1955), I personally wonder if the deflated Self does not risk becoming the essential prison of industrial society, in which, unfortunately, there is no Nirwana.... Professor Mourant's thirty pages of Introduction (supplemented appropriately by the great "Prayer" in which the Saint explains the world to his Creator), the bibliographical references, and the critical comments, all provide a concise exposition of the varioua ways in which Augustine attempted to organize his faith into a system of thought. The selections from the treatises, sermons, and letters are well arranged and contain several new translations into English by competent scholars. Both the editor's exposition and Augustine's own efforts to face the basic issues created by the meeting of diverse systems of doctrine and language enable the student to become aware of the amazing range of Christian philosophy created by a man who was both a busy bishop and an inquisitive truth-seeker. This paper is about the similarity between the appreciation of a piece of art, such as a cherished music album, and the loving appreciation of a person whom one knows well. In philosophical discussion about the rationality of love, the Qualities View (QV) says that love can be justified by reference to the qualities of the beloved. I argue that the oft-rehearsed trading-up objection fails to undermine the QV. The problems typically identified by the objection arise from the idea that (...) love-worthy qualities could be coarse-grained, when in fact they must be fine-grained. The analogy with appreciation of aesthetic qualities helps to draw out this point. Once the fine-grained nature of love-worthy qualities is properly understood, it is clear that critics of the QV cannot rely on the trading-up objection to motivate its rejection. Moreover, the paper’s core argument helps to illuminate the persistently aesthetic nature of interpersonal affections. (shrink)

A lineage of prominent philosophers who have discussed the second-person relation can be regarded as advancing structural accounts. They posit that the second-person relation effects one transformative change to the structure of practical reasoning. In this paper, I criticise this orthodoxy and offer an alternative, substantive account. That is, I argue that entering into second-personal relations with others does indeed affect one's practical reasoning, but it does this not by altering the structure of one's agential thought, but by changing what (...) reasons can become available. The importance of second-personal thought for action is heterogenous. Second-person relations make possible the emergence of a wide variety of different kinds of practical reasons: creating some, revealing others. Recognising this diminishes the appeal of the traditional, structural accounts of the practical significance of such second-personal relations. Moving away from structural accounts facilitates a more thorough understanding of the intersubjective form of action. (shrink)

Being able to ask others to do things, and thereby giving them reasons to do those things, is a prominent feature of our interpersonal lives. In this paper, I discuss the distinctive normative status of requests – what makes them different from commands and demands. I argue for a theory of this normative phenomenon which explains the sense in which the reasons presented in requests are a matter of discretion. This discretionary quality, I argue, is something that other theories cannot (...) accommodate, though it is a significant aspect of the relations that people stand in to one another, and the kinds of practical reasons that flow from those relations. For people to contribute to discourse, they must do more than utter the right sentence at the right time. The basic requirement is that they add to their common ground in an orderly way. To do this, we argue, they try to establish for each utterance the mutual belief that the addressees have understood what the speaker meant well enough for current purposes. This is accomplished by the collective actions of the current contributor and his or her partners, and these (...) result in units of conversation called contributions. Since a category is a type of concept, it appears from this account that Kant holds a linguistic theory of concepts in general. According to Bird, Kant identifies concepts with language (pp. 61, 121, 123-124); they are, for him, linguistic entities (pp. 100, 104). On one occasion he refers to Kant's theory as a "picture of language" (p. 102). Kant seems thus to (...) be transformed by Bird into an ordinary language nominalist--and that a century and a half before Wittgenstein. The trouble with all this is that it simply is not supported by the text. Rather than giving a linguistic interpretation of concepts in general and categories in particular, Kant constantly analyzes them in conceptual terms. He speaks of them as forms of thought, ideas, conditions or criteria of thought, acts of pure thought, and so on. Their origin is always the human understanding. But, as Bird states in the passage quoted, Kant believes that our language sets a limit to our understanding. This might be taken to mean that language controls our thought so that we can conceive only what we can verbally describe. Or it could have a stronger meaning--that thought is reducible to verbalization. Bird seems to accept the latter as the proper interpretation of Kant for he contends that Kant holds the view that to conceive something is to describe it linguistically (pp. 127-129). If these words are to be understood in their ordinary meanings, one seems forced to the conclusion that in Bird's mind Kant is a linguistic behaviorist. If this is what Bird means to imply, it hardly seems worthwhile to pursue his linguistic reformulation of the critical philosophy any further. Nevertheless, the question remains in the reader's mind, "Why should any student, particularly one who has shown such understanding and appreciation of Kant, depart from his scholarly approach to the Critique to attribute to Kant a theory which, instead of having any plausibility as an interpretation of the Kantian epistemology, seems rather to be a parody of the argument it is meant to explain?" The importance of Chauncey Wright in the history of American philosophy is being increasingly realized. Professor Madden's work is the first comprehensive account of his life, his writings, and his ideas. The sources are scanty and often obscure, but Madden has made exhaustive use of them and gives us a bibliography and critical notes. This is a product of first-class scholarship, bringing together what the author had published in scattered BOOK REVIEWS 263 articles, and supplementing his anthology of Wright (Liberal Arts Press). The biographical chapter presents Wright as an attractive character among devoted friends and also as a solitary, original scientist. Wright's primary achievement was to apply utilitarian principles to Darwinian natural selection theory. Since Darwin himself made no such attempt, nor did John Stuart Mill, and since Darwin showed an evident interest in Wright's attempt, this represents a major contribution to evolutionary theory. On the negative side, too, Wright was incisive in his criticisms of Spencer, Mivart, Hamilton, Mansel, and Lewes. The pragmatic strains in Wright's philosophical method were incidental to his primary interest in showing that sciences are metaphysically "neutral." But his influence on Peirce and James was direct and strong, both on their cosmological and methodological doctrines. He deserves to be mentioned as the prime mover in this trinity of early pragmatism and naturalism, but since his own writings were relatively few and scattered, his historical importance rests largely on the use which Peirce and James made of his ideas and the development which they gave to his philosophy. Clark highlights a neglected issue in research on language use: the process by which speakers and addressees anchor utterances with respect to individual entities in their common ground. In his review, he identifies the challenges linked to investigations of anchoring, but also displays the pitfalls of evading it.

Social robots serve people as tutors, caretakers, receptionists, companions, and other social agents. People know that the robots are mechanical artifacts, yet they interact with them as if they were actual agents. How is this possible? The proposal here is that people construe social robots not as social agents per se, but as depictions of social agents. They interpret them much as they interpret ventriloquist dummies, hand puppets, virtual assistants, and other interactive depictions of people and animals. Depictions as a (...) class consist of three physical scenes with part-by-part mappings between them: (a) a base scene (the raw physical artifact), (b) the depiction proper (the artifact construed as a depiction), and (c) the scene depicted (the scene people are to imagine). With social robots, evidence shows people form the same three scenes plus mappings: They perceive the raw machinery of a robot, construe it as a depiction of a character, and, using the depiction as a guide, engage in the pretense that they are interacting with the character depicted. With social robots, people also recognize three classes of agents – the characters depicted, the intended recipients of the depictions (those who view or interact with the robots), and the authorities responsible for the robots (the designers, makers, and owners). Construing social robots as depictions, we argue, accounts for many phenomena not covered by alternative models. (shrink)

In everyday joint activities, people coordinate with each other by means not only of linguistic signals, but also of material signals – signals in which they indicate things by deploying material objects, locations, or actions around them. Material signals fall into two main classes: directing-to and placing-for. In directing-to, people request addressees to direct their attention to objects, events, or themselves. In placing-for, people place objects, actions, or themselves in special sites for addressees to interpret. Both classes have many subtypes. (...) Features of material signals were examined in pairs of people who were videotaped as they assembled TV stands, built Lego models, planned furnishings for a house, played piano duets, or bought coffee at Starbucks. In these activities, the pointing and placements were often sustained, creating three phases of signals – initiation, maintenance, and termination – each with its own interpretation. (shrink)
